There's really no reason to think Sulu wasn't always a gay character. There's no recon here or anything.

My guess is that he considers himself an actor not a gay actor and feels bummed out that a character that wasn't written as gay has (maybe, probably) become gay because the actor who played him is a gay icon.

No he never had a love interest. His does have a daughter though. Which is why it doesn't contradict anything. Takei seems to feel that since Roddenberry didn't write him gay (mostly because it wouldn't have flown with the studio and the audience) its against Gene's vision. It's dumb but its how he feels.

Outside of Kirk, Star Trek characters are about as asexual as it gets. They could all be gay for all we know.

Yeah, there is actually quite a lot of sexuality in all incarnations of ST.
