Only one player can be in motion at the snap of the ball.
In the National Football League (NFL), an illegal shift occurs when an offensive player moves out of position before the ball is snapped without coming to a complete stop. The penalty for an illegal shift is a loss of five yards and a replay of the down.
Rules
The offensive team can shift players multiple times before the snap.
After the last shift, all players must stop and be in position for at least one second.
A player can be in motion before the snap, but two or more players in motion is an illegal shift.
The offensive team must have a legal formation before and after a shift.
The referee signals an illegal shift with both arms moved forward and level to about a 45 degree angle.
That blurb is a bit confusing. An "in motion" player is only considered "in motion" if they are still moving when the ball is snapped (hence why only one player can be in motion).
